Preferred Draw Volume

Lt Green 4.5 mL, Lt Blue 2.7 mL (x5), Lavender 3 mL (x2), Gold 5 mL

Unacceptable Conditions

Blue Tube: Whole blood or plasma on ice, clotted sample, sample collected in glass tube, unspun specimen > 4 hours old, underfilled tube, or gross hemolysis.
Gold Tube: Avoid hemolyzed and lipemic specimens.
Lt Green Tube: Samples thawed more than once or serum.
Lavender Tube: Clotted or contaminated sample or if collected in any other anticoagulant.

Collection Comments

Collect 1 Clear Discard, 5 Full 2.7 mL plastic Blue, 1 Gold, 1 Lt Green, and 2 Lavender in order left to right. Processing: Do not send whole blood or plasma to lab on ice. Double spin and freeze plasma within 4 hours of collection: Spin all five Blue tubes at 3000rpm for 15 min. Place top 2/3 of plasma in 5 aliquot tubes and spin aliquots at 3000 rpm for 15 min. Take top 2/3 of 2nd spin and put into 7 aliquot tubes. Mark aliquot tubes 'plasma'. Freeze all 7 aliquots immediately. Send 7 frozen aliquots of platelet poor plasma. Also send refrigerated RBC's from one Blue tube. Gold tubes - Spin and separate ASAP. No severely lipemic, contaminated, or hemolyzed specimens. Send 2 aliquots of refrigerated serum. Lt Green - Spin, separate and freeze plasma within 1 hour of collection. Mark tube 'plasma'. Send frozen plasma. Lavender - Do Not Spin. Send refrigerated whole blood. Remember to properly label the aliquot tube with patient name, DOB, test and collect date.

Careset includes: APC Resistance (APCR), PT Gene Mutation, AT III Functional, Factor VIII Assay, Protein C Activity and Protein S Antigen Free (if patient is not on Coumadin), Homocysteine, Lupus Anticoagulant (DRVVT and StaClot LA), Cardiolipin Ab IgG/M, Beta 2 Glycoprotein 1 IgG/M Ab. Reflex testing for Factor V Leiden PCR will be performed if APCR is abnormal. A MTHFR Mutation Bld will be performed if homocysteine is abnormal.
