Minimum Volume: 0.5 mL Serum

Specimen Preparation

Plain Red or Serum Separator Tube (SST)
  • Allow sample to clot fully for 30 minutes at room temperature
  • Centrifuge within 2 hours of collection
  • Transfer serum to plastic transport tube.

Samples may be frozen and thawed up to 3 times.  Mix frozen samples well after thawing.

Storage/Transport Temperature

Frozen.  Submit specimen in plastic transport tube.

Stability (from collection to initiation)

Ambient: 24 hours
Refrigerated: 7 days
Frozen: 1 month

Unacceptable Conditions

Hemolyzed, icteric, lipemic or bacterially contaminated sera.

Methodology

Indirect Immunofluorescence using Hep-2 cells

Interpretive Data

ANA is a standard screening assay for detecting multiple autoantibodies produced by patients with an ANA associated rheumatic disease.  In 2009 a task force of the American College of Rheumatology issued a statement declaring Hep-2 indirect immunofluorescence as the preferred method for ANA screening.

Avera McKennan Laboratory will report 5 patterns and respective titers: Homogeneous, Speckled, Centromere, Nucleolar and Nuclear Dots.  Cytoplasmic staining will be noted if present.
